User Guide

The BMRZ offers a comprehensive RNA production service designed to support high-quality RNA sample preparation for liquid-state NMR spectroscopy and related structural biology applications.

Our facility provides expertise in biochemical RNA synthesis and labeling, including:

  • Selective deuteration to enhance NMR spectral quality,
  • 15N and/or ¹³C isotope enrichment (uniform or nucleotide specific) for multidimensional NMR experiments, and
  • Segmental labeling using one-step ligation of in vitro transcribed RNA fragments, allowing targeted isotope incorporation into specific RNA regions.

These approaches enable detailed structural and dynamic studies of RNA molecules by NMR, improving both spectral resolution and interpretability.

Researchers planning to use the RNA production service are strongly encouraged to review the following publications for background information and methodological insights:

  • For an overview of RNA NMR spectroscopy: DOI: 10.1002/cbic.200300700
  • For detailed information on RNA preparation requirements and practical tips: https://doi.org/10.1002/cpnc.116

For project-specific discussions, including design of labeling strategies, RNA construct planning, and production feasibility, please contact the Facility staff: Harald Schwalbe and Christian Richter

They will provide guidance to ensure that RNA samples meet the necessary standards for successful NMR-based structural studies.
