CCP4 Cloud for structure solution - IGBMC

Meeting
Date: 26-Sep-2023

Instruct Partner

The workshop will provide an overview of the development of structural solution projects using CCP4 Cloud, a modern interface for the CCP4 Software Suite. CCP4 Cloud includes tasks for all stages of computational structure determination and provides seamless access to relevant web resources such as PDB, UniProt, AlphaFold, ESM, and third party software such as Global Phasing's Buster and NKI's PDB-REDO, Amsterdam. It offers a comprehensive suite of automated tools for image processing, phasing, model building, refinement, validation, and preparation for deposition in the PDB. The hallmark of CCP4 Cloud is its ability to automatically develop structure solution projects capable of delivering complete solutions, while allowing user intervention when a satisfactory result is not achieved.

During this workshop, attendees will be introduced to the advanced features of CCP4 Cloud, learning the powerful tools and workflows available for structure determination. We will focus on advanced techniques such as Molecular Replacement using AlphaFold-2 models, automated approaches to structural solution and project development.

One of the key strengths of CCP4 Cloud is the facilitationof collaborative research and education. We'll demonstrate how CCP4 Cloud allows multiple individualsto work on shared projects simultaneously, in real time. Next, we will introduce CCP4 Cloud Archives, a new feature that allows researchers to store completed projects in the Cloud and make them available via stable web links similar to DOI. Project archiving facilitates publishing, peer review, learning, and knowledge sharing in a broader collaboration. It also facilitates data retention in structural biology by bridging the gap between experimental observations and their interpretations in the form of solved structures deposited in the PDB.

This workshop is specifically designed to meet the needs of a diverse audience, including PhD students, postdoctoral researchers, and Principal Investigators in the field of structural biology who are enthusiastic about adopting the latest crystallography software from CCP4 into their work. The workshop will begin with an introductory lecture on CCP4 Cloud, which will provide an overview of its features, capabilities and benefits in structure determination. Participants will gain a comprehensive understanding of the software and how to solve structures with it.

After the introductory lecture, the workshop will move into a hands-on session where participants can learn about CCP4Cloud through tutorials or develop their own structural solution projects. The emphasis of the workshop will be on modern approaches to structural solution, in particular on Molecular Replacement using AlphaFold. The workshop will help participants make the most effective use of the CCP4 software in their research.
